- [00:00:10.350]We are located in North Wing
- [00:00:11.660]of the Morrison Life Science Research Center

- [00:00:17.890]The laboratory is large and easy to social distance
- [00:00:20.400]and we have open bench space available
- [00:00:22.050]and a centrifuge for use if users wish
- [00:00:24.610]to prepare their samples in the lab.
- [00:00:26.810]The center is open for business Monday through Friday
- [00:00:28.980]from 8:00 to 5:00,
- [00:00:30.340]evening and weekend access may be granted to users
- [00:00:32.460]with appropriate training.
- [00:00:34.600]At the moment, the center has two primary instruments
- [00:00:36.820]to meet researchers needs.
- [00:00:38.270]The newest addition to the facility
- [00:00:39.680]is the Beckman Coulter CytoFLEX LX analysis system.
- [00:00:43.030]The CytoFLEX is equipped with four laser lines
- [00:00:45.500]and 16 fluorescence detectors.
- [00:00:47.920]It is driven by the intuitive site expert software
- [00:00:50.760]and I do offer training on this system
- [00:00:52.220]if users are interested in running their own experiments
- [00:00:54.670]or I will be happy to run experiments for customers.
- [00:00:57.470]In addition to the fluorescent detectors,
- [00:00:59.250]it is also equipped with extremely sensitive
- [00:01:01.160]forward and side scatter detectors
- [00:01:02.960]that are capable of visualizing particles
- [00:01:04.720]as small as 300 nanometers.
- [00:01:06.680]So it is capable of analyzing extracellular vesicles
- [00:01:10.120]and even other nanoparticles.
- [00:01:12.770]Most of the work that is done on the system
- [00:01:14.330]is looking at multi-parameter fluorescence panels
- [00:01:16.860]with an option to load samples into 96-well plate
- [00:01:19.430]for automatic acquisition.
- [00:01:21.100]It has proven to be the lab's workhorse.
- [00:01:23.690]The other main system in the lab
- [00:01:25.100]is our BD FACSAria II cell sorter.
- [00:01:27.510]This system also has four laser lines working
- [00:01:29.720]with 17 fluorescence detectors
- [00:01:31.760]and can be used for analysis if necessary.
- [00:01:34.110]But the real benefit of this system
- [00:01:35.580]is that it is capable of collecting
- [00:01:37.030]and purifying specific subsets from the sample.
- [00:01:39.960]For example, if you are working with a transfected cell line
- [00:01:42.940]and you only want to work with the GFP-positive cells,
- [00:01:45.810]we can run the sample through this order
- [00:01:47.240]and collect and isolate only the cells
- [00:01:49.000]that have expression of GFP
- [00:01:51.040]while removing all the negatives.
- [00:01:53.180]You could also sort your GFP containing cells directly
- [00:01:55.610]into a multi-well plate
- [00:01:56.830]as few as one cells per well if needed
- [00:01:59.780]and put them back into culture to grow further.
- [00:02:02.160]The sorter is housed in the biosafety cabinet
- [00:02:04.140]for user and staff protection
- [00:02:05.770]and because of that,
- [00:02:06.603]we were able to work with BSL-2 Plus level samples.

- [00:02:11.870]to the university to come and use our facilities.
