Designing the gene is the first step of genetic engineering. This video provides a brief summary of plant gene design as part of the SDS in soybean discussion in Journey of a Gene.

[00:00:02.210]of genetic engineering, which is designing the gene.
[00:00:06.410]So the goal of this step was to modify
[00:00:09.000]the Arabidopsis SDS resistance gene
[00:00:11.890]so it will work in soybeans.
[00:00:14.630]So this had three steps to it: extract, copy, and modify.
[00:00:22.910]With extracting, that means you're taking all the DNA
[00:00:26.150]out of the Arabidopsis plant
[00:00:27.820]and then you're also taking DNA out of a soybean plant.
[00:00:32.500]Then what you do is you make copies
[00:00:34.480]and you're copying just the region
[00:00:36.140]that you want as a piece of your gene.
[00:00:39.060]So for the Arabidopsis plant,
[00:00:41.320]we're just copying this coding region.
[00:00:43.990]This is the part of the gene that codes to make the protein
[00:00:48.810]for resistance to the disease.
[00:00:50.960]So this is the functional part.

[00:00:54.710]to know when to turn the gene on,
[00:00:56.420]it needs that soybean promoter.
[00:00:58.830]So this promoter is the on/off switch for the gene
[00:01:02.500]and so we need to make copies of that soybean promoter
[00:01:07.150]and then stick them together to make the transgene.
[00:01:09.750]So this has the soybean on/off switch.
[00:01:12.080]So the soybean plant knows when and where
[00:01:14.240]to turn on this gene
[00:01:15.580]and then that Arabidopsis disease resistance gene
[00:01:18.890]that the soybean needs in order to not be affected

[00:01:27.630]So then from there, Midan will take this new transgene
[00:01:32.010]and pass it on to Shirley
[00:01:33.470]and what she'll do is take the transgene
[00:01:35.300]and put it into soybean plants,
[00:01:37.600]which can later be bred and tested
[00:01:40.180]to finish off the process.
